摘要
目的观察红花黄色素对缺氧后脑内诱生型一氧化氮合酶(iNOS)、神经原型一氧化氮合酶(nNOS)及内皮型一氧化氮合酶(eNOS)基因表达的影响,探讨红花黄色素抗缺氧脑损伤的作用。方法采用SD新生鼠缺氧模型,于缺氧前30min腹腔注射红花黄色素生药7g/kg,缺氧40min后复氧48h,提取脑组织总RNA,应用RTPCR技术检测三种NOSmRNA的表达量。结果新生鼠缺氧再复氧48h,脑内iNOS、nNOS基因表达上升(P<0.05),预先给予红花黄色素能抑制iNOS、nNOS基因的表达(P<0.05),但eNOS基因表达不受影响。结论红花黄色素对缺氧脑损伤的保护作用与NOS基因表达有关。
Aim: To investigate the effect of Safflor yellow on the gene expression of neuronal nitric oxide synthase(nNOS),inducible nitric oxide synthase(iNOS) and endothelial nitric oxide synthase(eNOS) in neonatal asphyxia. Methods: 30 minutes after SY 7 g/kg weight intraperitoneally was administered on the neonatal rats. After asphyxia for 40 minutes,the neonatal rats were reoxygenated for 48 h,and the nitric oxide synthases(NOSs) mRNA expression was assessed by semi quantitative reverse transciption polymerase chain reaction. Results: Neuronal nitric oxide synthase(nNOS) and inducible nitric oxide synthase(iNOS) were up in hypoxia/reoxigenation(H/R)48 h gounp,while both of them were down significantly in SY gounp,but no chang was observed on endothelial nitric oxide synthase (eNOS). Conclusion: The protective of SY from brain damage induced by neonatal asphyxia might be associated with expression of NOSs mRNA.
