青蒿素抗柯萨奇B组病毒感染的实验观察 被引量:10

摘要 目的探讨青蒿素体外抗柯萨奇B组3型病毒(CVB3)的作用。方法以CVB3感染HeLa细胞为实验模型,采用微量细胞病变抑制法、半定量RT鄄PCR实验和间接免疫荧光实验的方法,观察分析青蒿素对CVB3的直接灭活、阻断吸附和抑制复制的作用,以及青蒿素对CVB3在感染细胞内核酸复制和蛋白表达的影响。结果青蒿素不同程度地阻断病毒吸附和抑制病毒复制,明显抑制CVB3核酸复制与蛋白表达,未发现有直接灭活病毒的作用。结论青蒿素类药物具有体外抗CVB3的作用,其抗病毒机制是通过阻断病毒吸附和抑制病毒复制来完成的。 Objective To investigate the anti-Coxsackie virus B3 (CVB3) effect of artemisinin in vitro. Method The anti-CVB3 effect of artemisinin was detected by micro cytopathogenic effect inhibition assay in vitro. Moreover, semi-quantitive RT-PCR and indirect immunofluorescence assay were used to investigate the inhibitive effect of artemisinin on CVB3 RNA replication and protein expression. Results Artemisinin had inhibition on CVB3 adherence and replication to different extents according to the ever increasing drug concentration. Artemisinin(15.0 μg/ml) efficiently inhibited CVB3 RNA replication and protein expression. Direct virucidal activity of artemisinin was not found. Conclusions Artemisinin plays an important role in the anti-CVB3 effect in vitro by means of inhibiting CVB3 from adhering to the host cells and replicating in them.
