摘要
目的 建立稳定的肝移植大鼠全肠外营养 (TPN)模型。方法 以二袖套法进行大鼠原位肝移植 ,并采用改制的代谢笼及输液保护架 ,术后 6h从右颈外静脉给予含中长链脂肪乳剂的TPN ,观察其大体情况及 7d存活率、肝功能、移植肝组织学改变。结果 3 0例肝移植大鼠TPN支持后 7d存活率 86 7% ( 2 6 3 0 ) ,光镜检查肝细胞无坏死 ,电镜检查线粒体结构完整。结论 本实验所建立的大鼠肝移植TPN模型稳定、重复性好、易于操作。
Objective To establish a stable model of total parenteral nutrition (TPN) in rats with liver transplantation. Methods Orthotopic liver transplantation was performed and the portal vein and hepatic inferior vena cava were reconstructed by the cuff technique in rats. TPN was given with modified tube and retainer at 6 h after operation. The 7-day survival rate, liver function, and pathological changes of the liver were observed. Results The animal survival rate was 86.7% (26/30). Light microscopy and electron microscopy showed that the morphology of hepatocytes and mitochondrion was normal after TPN for 7 d. Conclusion The model of TPN in rats with liver transplantation established in this study is stable, reproducible, and easy to operate.
