摘要 目的 :建立HPLC法同时测定人血浆中地西泮、硝西泮、氯硝西泮、咪达唑仑、三唑仑、艾司唑仑和阿普唑仑的浓度。方法 :血浆样品用乙醚萃取 ,5 5℃水浴中N2 吹干 ,残留物用乙醇溶解后进样。色谱柱为Shim packCLC CN柱 (15 0mm× 6 0mm ,5 μm) ,流动相为正己烷 -无水乙醇 -甲醇 (87 8∶10 2∶2 ,V/V/V) ,流速 1 10mL·min-1,柱温 4 0℃ ,检测波长 2 4 0nm。结果 :硝西泮、氯硝西泮在 0 0 2~ 2 0 0mg·L-1范围内线性关系良好 (r≥ 0 9993) ,地西泮、咪达唑仑、三唑仑、艾司唑仑和阿普唑仑在 0 0 5~ 2 0 0mg·L-1范围内线性关系良好 (r≥ 0 .9997)。方法回收率近 10 0 % ,日内和日间RSD均≤ 7% (n =5 )。结论 :本方法适用于人血浆中 7种苯二氮 艹卓 AIM: To establish a HPLC method for simultaneous determination frequently prescribed 7 benzodia-zepines: diazepam,nitrazepam,clonazepam, midazolam, estazolam,triazolam and alprazolam in human plasma. METHODS: The plasma samples were extracted with ether. The organic phase was evaporated under nitrogen stream at 55 ℃ in water bath. The remains were dissolved by absolute ethanol. Column: Shim-pack CLC-CN (150 mm×6.0 mm,5 μm); Mobile phase: n-hexane-absolute ethanol-methanol (87.8∶10.2∶2,V/V/V) at a flow rate of 1.10 mL·min -1. The column temperature was 40 ℃, ultraviolet detection wavelength was at 240 nm. RESULTS: The linear ranges were 0.02-20.0 mg·L -1(r≥0.999 3) for nitrazepam and clonazepam. The other linear ranges were 0.05-20.0 mg·L -1 for diazepam,midazolam, estazolam, triazolam and alprazolam(r≥0.999 7). The average recovery of 7 drugs were nearly 100%. Intra-day RSD and inter-day RSD were less than 7%(n=5). CONCLUSION: The method is suitable for simultaneous determination of diazepam, nitrazepam, clonazepam, midazolam, estazolam,triazolam and alprazolam in human plasma.
