摘要
目的 :研究大孔吸附树脂纯化远志总皂苷的工艺条件及参数。方法 :以远志总皂苷的洗脱率、精制度为指标 ,考察大孔吸附树脂对远志总皂苷的吸附性能和洗脱参数。结果 :12mL远志总皂苷样品液 (生药 0 2 5g/mL)上大孔吸附树脂柱 (Φ15mm×H90mm ,干重 2 5g) ,用蒸馏水、30 %乙醇、70 %乙醇各 3BV依次洗脱 ,远志总皂苷富集于 70 %乙醇洗脱液部位。结论 :通过大孔吸附树脂富集与纯化 ,远志总皂苷洗脱率为 89 8% ,精制度为 2 39 6 %。
Objectives: To study the technological parameters of purifying saponin with macroporous resin. Method: With the elution rate and purity as indexes, the absorption characteristics and elution parameters were studied. Result: 12 mL of the extractive of Radix Polygalae (crude drugs 0.25g/mL) was added on a column of macroporous resin (Φ15mm×H 90mm, dry weight 2.5g) and was washed with 3BV of distilled water, 30% ethanol and 70% ethanol, successively. Saponin was enriched in the 70% ethanol elution. Conclusion:Enriched and purified by macroporous resin, the elution rate of saponin was 89.8% and the purity reached 239.6%. So, the established method is feasible.
