摘要
研究了高效液相色谱法测定板栗内源激素——赤霉素(GA3)、3-吲哚乙酸(IAA)、玉米素(Z)和脱落酸(ABA)的条件;采用WatersC18反相柱(4.6mmi.d.×250mm,5μm),SPD-6AV紫外检测器,以甲醇-水-乙酸(体积比为45.0∶54.2∶0.8)溶液为流动相,流速为1.0mL/min,进样量为20μL,检测波长为254nm,选用外标法进行了定量测定.试验结果表明:其回收率高,GA3、IAA、Z、ABA的检出限分别为0.50、0.10、0.30和0.03mg/L.
A simple, rapid and perfect extraction and determination method for the internal hormones [gibberellin acid (GA_3), indole-3-acetic acid (IAA), zeatin (Z), abscisic acid (ABA)] in chestnuts with phase-reversed, high-efficient liquid chromatography has been developed. The chromatographic conditions are as follows: a waters C_(18) phase-reversed column (4.6 mm i.d.×250 mm, 5 μm), an SPD-6AV detector, and methanol-water- acetic acid liquid (volume ratio: 45.0∶54.2∶0.8)as the mobile phase at the velocity of 1.0 mL/min with an input of 20 μL and a wavelength of 254 nm. The test results show that the rate of recovery is high and the detection limits are 0.5 mg/L for GA_3, 0.1 mg/L for IAA, 0.3 mg/L for Z and 0.03 mg/L for ABA.
