复方水蛭合剂对大鼠局灶性脑缺血细胞因子免疫损伤的保护作用

Protecting effect of Complex Hirudo on immunity injury of cytokine in rats with focal cerebral ischemia

目的:探讨复方水蛭合剂对局灶性脑缺血细胞因子免疫损伤的保护作用。方法:Wistar大鼠正常组6只,空白对照组12只,复方水蛭合剂组36只。检测大鼠大脑中动脉缺血后6,24h脑组织匀浆白细胞介素1、肿瘤坏死因子、一氧化氮的变化及复方水蛭合剂对其影响。结果:大鼠大脑中动脉缺血后6,24h模型组脑组织匀浆肿瘤坏死因子、白细胞介素1、一氧化氮犤6h:(73.47±7.04)kIU/L,(0.87±0.05)μg/L,(89.07±6.69)μmol/L;24h:(90.75±7.63)kIU/L,(0.98±0.07)μg/L,(114.16±9.03)μmol/L犦明显比正常组犤(57.55±4.85)kIU/L,(0.70±0.09)μg/L,(51.43±5.49)μmol/L犦高(P<0.01),大、中剂量复方水蛭合剂组明显比空白对照组低(P<0.05～0.01,)。病理形态学检查:大、中剂量复方水蛭合剂组与空白对照组比较均较轻。结论:复方水蛭合剂对脑缺血所致的细胞因子的免疫损伤有保护作用。

AIM:To investigate the protective role of Complex Hirudo in cytokine immunity injury of rats following focal cerebral ischemia. METHODS:Contents of interleukin-1(IL-1),tumor necrosis factor(TNF),and nitri c oxide(NO) were measured 6 and 24 hours after middle cerebral artery(MCA) ische mia in the rats of normal control group(n=6),blank control group(n=12) and Compl ex Hirudo group(n=36). RESULTS:Six or twenty-four hour after MCA ischemia,the contents of IL-1, TNF and NO in the blank control group[6 hours:(73.47±7.04) kIU/L,(0.87±0.05)μg/L ,(89.07±6.69)μmol/L;24 hours:(90.75±7.63) kIU/L,(0.98±0.07)μg/L,(114.16±9. 03) μmol/L] were significantly higher than those of the normal control group[(5 7.55±4.85) kIU/L,(0.70±0.09) μg/L,(51.43±5.49) μmol/L](P< 0.01),and those o f the large-dosage and middle-dosage Complex Hirudo groups(P< 0.05-0.01).The pathological injury in large and middle dosage Complex Hirudo groups was milder than that in the blank control group. CONCLUSION:Complex Hirudo is effective for protecting rats against cytokine im mune injury induced by cerebral ischamia.
