摘要 利用Chiralpak AD-H手性柱建立了氢溴酸西酞普兰对映体HPLC手性拆分的方法,同时对草酸依地普仑(即右旋西酞普兰)进行了ee值检查。流动相为正己烷-异丙醇-二乙胺(95∶5∶0.1),柱温25℃,检测波长240nm。S-(+)-及R-(-)-对映体的分离度为2.47。并测得草酸依地普仑中R-(-)-西酞普兰含量小于1.0%,S-(+)-西酞普兰的ee值大于98.0%。 A HPLC method for chiral separation of citalopram hydrobromide enantiomers and optical puritydetection of escitalopram oxalate was established. A Chiralpak AD-H chiral column was used with the mobile phase ofn-hexane-isopropylalcohol-diethylamine(95∶5∶0.1). The column temperature was 25℃ and the detection wavelengthwas 240nm. The average resolution between S-(+)- and R-(-)-citalopram was 2.47. The R-(-)-citalopram contentwas less than 1.0%. The ee of escitalopram oxalate was more than 98.0%.
