摘要
目的 :探讨外源性雌激素在脂多糖 (LPS)诱导大鼠急性肺损伤 (ALI)中对肺脏病理改变及作用机理。方法 :选用Wistar大鼠 60只 ,随机分成 3组 :正常对照组 (静注生理盐水 )、LPS组 (尾静脉注射LPS 5mg/kg)和LPS +雌二醇组 (注射LPS 5mg/kg +雌二醇 1mg/kg) ,2 4h后观察大鼠肺脏病理变化 ,支气管肺泡灌洗液 (BALF)中蛋白含量 ,肺通透指数 ,外周血中雌二醇和催乳素浓度 ,肺组织中雌激素受体、孕激素受体表达。结果 :注射LPS后肺泡灌洗液中蛋白含量及肺通透指数均升高 (p <0 0 5 ) ,给予外源性雌激素可显著减轻上述变化 (p <0 0 5 )。LPS +雌二醇组外周血中雌二醇和催乳素浓度 ,肺组织中雌激素受体、孕激素受体含量较LPS组有显著性差异 ,肺组织损伤较轻。结论 :外源性雌激素对脂多糖诱导的急性肺损伤有保护作用。
Objective: To explore the pulmonary pathological changes and mechanism of extra estrogen in acute lung injury induced by lipopolysaccharide. Methods: 60 Wistar rats were divided randomly into three groups, i.e. control group(injected saline), LPS group(injected endotoxin 5 mg/kg) and LPS+E group(injected endotoxin 5 mg/kg and estradiol 1 mg/kg). After injection 24 h, the content of proteins in bronchoalveolar lavage fluid(BALF), pulmonary permeability index and levels of estradiol and prolactin in rats' peripheral blood were examined, and the expressions of estrogen and progesterone receptors in the lung tissues were observed in the animals with acute lung injury induced by lipopolysaccharide. Results: In LPS group, the protein content in BALF and pulmonary permeability index increased obviously(p<0.05), but these increases would be attenuated by extra estrogen(p<0.05). There were significant differences between LPS group and LPS+E group in the levels of estradiol and progestogen in the peripheral blood, and the expression of estradiol and progestogen receptors in the animals' lung tissues. Among three groups, the lung injury of LPS+E group was the minimal. Conclusion: Extra estrogen could prevention of acute lung injury induced by lipopolysaccharide.
