摘要 观察了陡脉冲对恶性肿瘤细胞生长抑制作用及其细胞周期的影响。人卵巢腺癌 SKOV3细胞受不同剂量的陡脉冲作用后 ,分别制作细胞生长曲线和应用流式细胞术检测肿瘤细胞增殖及其对细胞周期的影响。经陡脉冲作用后的癌细胞的生长受到不同程度的抑制 ,随着陡脉冲剂量的增加 ,癌细胞的死亡率和抑制效应更加显著 ;陡脉冲选择性作用于肿瘤细胞 DNA合成、分裂期 ,出现 G0 / G1 期阻滞 ,抑制肿瘤细胞的增殖 ,陡脉冲剂量较大的实验组和对照组间比较有显著性 (P<0 .0 1)。陡脉冲对恶性肿瘤细胞具有杀伤和抑制作用 ,为其用于肿瘤治疗提供依据。 To assess study the cytocidal and inhibitory effects of steep pulsed electric fields (SPEFs) on ovarian cancer cell line SKOV3,the cancer cell suspension was treated by SPEFs with different parameters (frequency,pulse duration,peak value of voltage). Viability rate and growth curves of two test groups(high dosage and low dosage of SPEFs)and one control group were also measured. The DNA contents and cell cycle were analyzed by flow cytometry (FCM). Different dosing levels of SPEFs exerted obviously different effects on cancer cell viability. With the enhancement of each pulse parameter,the viability rate was promoted and the inhibitory effect on the proliferation of treated cells was more evident. The cells exposed to SPEFs grew slower than the control. The ratio of S+G 2/M phase cells was decreased,which restrained the DNA synthesis and division,but the ratio of G 0/G 1 phase cells was increased in the treated groups. It was also indicated that the SPEFs blocked the cell transition from G 0/G 1 phase to S+G 2/M phase. There was a significant difference in cell cycle between treated group and control group (P<0.01). Lethal effects of SPEFs were represented by inhibiting the cancer cell proliferation at the cell level and by influencing the cell cycle at the DNA level.
