摘要  研究了用微柱高效液相色谱法测定葛根中的葛根素的方法。葛根样品中的葛根素用甲醇超声振荡提取,然后以WatersXterraTMRP18(1.0×50mm,2.5μm)微柱为固定相,35%的甲醇(内含1%的醋酸)为流动相分离,在该色谱条件下,葛根素在2.0min内可达到基线分离;用紫外二极管矩阵检测器检测。方法标准回收率为97%~103%,相对标准偏差为1.6%~2.1%。方法测定了几种葛根样品中的葛根素,结果满意。 Microcolumn high performance liquid chromatography (HPLC) method for the determination of puerarin from Pueraria lobata was studied. Puerarin was extracted from sample with methanol by ultrasonic extraction for 45?min, separated on a Waters XterraTM RP18 (1.0×50?mm, 2.5?m) chromatographic microcolumn eluting with 35% of methanol containing 1% of acetic acid at a flow-rate of 0.2?ml/min monitored with a photodiode array detector at 250?nm. Recoveries of the flavonoid were 97 ~103% and relative standard deviations were 1.6 ~2.1%. The method was applied to the determination of puerarin in Pueraria lobata with good results.
