摘要
目的 探讨 MMP- 2 / 9与脑出血后脑水肿的关系。方法 采用胶原酶复制大鼠脑出血模型 ,通过免疫组化和原位杂交技术测定血肿周围脑组织中 MMP- 2 / 9的 m RNA和蛋白表达 ,同时进行脑水含量和渗出脑血管外 EB量的测定。结果 大鼠脑出血后血肿周围有明显的脑水肿发生和 EB含量显著增加 ,均于 2 4~ 4 8h达高峰。正常和假手术组可见极少量 MMP- 2蛋白和 m RNA阳性染色细胞 ;而无 MMP- 9的表达。模型组出现大量 MMP- 2 / 9蛋白和 m RNA阳性染色细胞 ;且 MMP- 9蛋白和 m RNA表达在 2 4~ 4 8h达高峰 ,与脑水含量和 EB含量呈正相关。结论 大鼠脑血肿周围早期存在 MMP- 2 / 9的表达上调 。
Objective To study the relationship of MMP-2/9 to the brainedema formation following ICH.Methods The ICH animal models wereestablished by stereotaxical injecting 0.4IU Ⅶ collagenase into the right globes pallidus(GP).The expression of MMP-2/9 protein and mRNA in the ICH rat brains had been investigated by immunohistochemistry and in situ hybridization.Brain water content(BWC)and EB contents were measured. Results Afteroperating 24~48h,we found that the exosmose dose of BWC and EB in perihematom(right side) at each time point was much higher than the left side and the right side of sham-operated group(SG).We found few MMP-2 protein and mRNA positivecells in the brain of NG and SG,and no MMP-9 protein and mRNA positive cells.There were lots of deep buffy positive cell of MMP-2/9 at each time point of operation group.The expression of MMP-9 protein and mRNA positive cells reached the peak at 24~48h in the operation group. There was positive correlation betweenthe expression of MMP-9 and mRNA positive cells and BWC and EB contents. Conclusion The expression of MMP-2/9 in perihematom were increasedat the early of ICH in rat,which may be correlated with forming the angiosoure encephaledema.
