摘要
目的 观察杏仁核点燃癫痫鼠模型脑内不同核团 FOS蛋白表达顺序及强度以探讨癫痫的传播途径。方法 选择健康 Wistar大鼠 6 0只制作杏仁核点燃癫痫模型 ,分别在点燃后 0 .5 h、1h、2 h、4 h及 2 4 h用免疫组织化学方法观察癫痫大鼠脑内梨状皮质、杏仁核、海马、齿状回、尾壳核、皮质、皮质下结构、小脑 Purkinje细胞、小脑齿状核、脑桥网状结构共 10个核团 FOS蛋白表达情况。结果 杏仁核点燃后 0 .5 h梨状皮质、齿状回、小脑 Purkinje细胞开始出现 FOS表达 ,然后表达逐渐增强 ,杏仁核、海马、小脑齿状核、皮质、皮质下结构、尾状核及脑桥网状核在发作后 1~ 2 h开始表达 ,4 h表达明显 ,癫痫发作 2 4 h后大多数核团 FOS蛋白表达低于 4 h但仍高于 1h~ 2 h。结论 杏仁核点燃大鼠癫痫发作后可引起脑内 FOS蛋白区域性一过性表达。梨状皮质表达最早 ,是首先被激活的结构之一 ,而杏仁核、海马、小脑 Purkinje细胞。
Objective To explore the transmission route of epileptic seizures by observing the Fos-like protein expression in nucleus of amygdaloid kindling rat model.Methods Sixty healthy Wistar rats wereselected to prepare the amygdaloid kindling model.The Fos-like protein expression at,1,2,4 and 24h after kindling was observed by immunohistochemical method in piriform cortex amydala,hippocampus,dentate gyrus,caudatus,putamen nuclei,cortex,subcortical structure,cerebellar Purkinje cells,cerebellar dentate nuclei,and pontine reticular nuclei,respectively.Results The Fos-likeprotein expressed firstly in piriform cortex,dentate gyrus and cerebellar Purkinje cells half an hour after kindling, and then the expression intensifiedgradually.The Fos-like protein began to express in amydala,hippocampus,cerebellar dentate nuclei,cortex,subcortical structure,caudatus putamen nuclei and pontine reticular nuclei from 1 to 2h after kindling,and the expression reached the peak in4h after kindling.Conclusions Amydala kindling seizure might induce regional and transient Fos-like protein expression in the rat brain.The piriform cortex might be the earliest region of Fos-like protein expression andone of the first activated structure.The amydala,hippocampus,cerebellar Purkinjecells and the cortex might be the important parts of the transmission route inamygdaloid kindling model.
