Survivin基因在胰腺癌组织中的表达及与bcl-2和p53蛋白表达的关系 被引量:2

Expression of Survivin gene and its relationship with expression of bcl-2,p53 proteins in pancreatic carcinoma

摘要 目的探讨Survivin基因在胰腺癌中的表达及与bcl 2、p5 3蛋白表达的相互关系。方法用RT PCR法检测 4 9例胰腺癌、5例慢性胰腺炎和 12例正常胰腺组织中SurvivinmRNA的表达 ,免疫组织化学法检测胰腺癌中bcl 2、p5 3蛋白表达。结果SurvivinmRNA在胰腺癌中表达率为76 % ,在慢性胰腺炎及正常胰腺组织中不表达 (P <0 0 5 )。Survivin基因表达与胰腺癌的分化程度、临床分期及淋巴结转移无相关性 (P >0 0 5 )。Survivin基因表达与bcl 2蛋白表达不相关 ,与p5 3蛋白表达显著相关。结论Survivin基因在胰腺癌中过度表达 ,提示该基因对胰腺癌发生和发展起重要作用 ;抑癌基因p5 3的失活与Survivin基因的表达可能在胰腺癌的发生中起协同作用。 Objective To study the expression of Survivin gene and its relationship with the expression of bcl-2,p53 in pancreatic carcinoma (PC). Methods[WT5”BZ] The expression of Survivin mRNA was investigated by reverse transcriptase polymerase chain reaction (RT-PCR) in PC samples of 49 cases ,chronic pancreatitis (CP) samples of 5 cases,normal pancreatic tissue samples of 12 cases. The expression of bcl-2 and p53 was detected by immunohistochemical assay. Results The expression rate of Survivin gene increased significantly in PC (75.5%) than in CP (0/5) and normal pancreatic tissue ( 0/12 ) ( P <0.05). There was no relationship between Survivin gene expression and histological differentiation,clinical stages,or lymph node metastasis of PDC. The expression of Survivin gene correlated with p53 expression,but not bcl-2 expression. Conclusions Survivin may play an important role in pathogenesis of PC. Up-regulated Survivin and disfunction of p53 may play a synergetic role in the process of carcinogenesis of PC.
