乐果接触工人外周血淋巴细胞M_3受体mRNA水平的研究 被引量:4

摘要 目的 研究乐果接触工人外周血淋巴细胞毒蕈碱型乙酰胆碱受体 (M3)基因表达情况 ,初步探讨M3 受体mRNA水平在有机磷农药毒性效应中起的作用。方法 选取乐果接触工人 3 3人和对照人群 15人 ,分离其外周血淋巴细胞后体外用乐果处理 ,用逆转录聚合酶链反应 (RT PCR)法扩增其M3 受体mRNA ,计算mRNA基础水平和诱导水平。结果 乐果接触工人和对照人群外周血淋巴细胞M3 受体mRNA基础水平相近 ,相对值分别为 (1.49± 0 .2 0 )和 (1.49± 0 .45)。经体外乐果诱导后 ,乐果接触组mRNA水平为 1.92± 1.0 7,明显高于对照组 (1.2 2± 0 .19)与自身基础水平 (1.49± 0 .45) ,差异均有显著性 (P <0 .0 5)。两组被测人群外周血淋巴细胞M3 受体mRNA基础水平和诱导水平均无性别差异。乐果接触人群M3 受体mRNA的诱导水平随接触工龄的增加而升高 ,工龄 <5年者为 (1.69± 0 .95) ,5~ 2 5年者为 (1.91± 1.0 3 ) ,>2 5年者为 (2 .0 9± 1.2 5) [与工龄 <5年者差异有显著性 (P <0 .0 5) ]。结论 长期接触乐果后 ,工人外周血淋巴细胞M3 受体mRNA基础水平与对照人群差异不明显 ,但体外诱导后其M3 受体mRNA诱导水平明显上升 。 ObjectiveTo study the expression of muscarinic receptor M 3 gene in peripheral blood lymphocytes of workers exposed to organophosphorus pesticides(OPPs) and to explore its role in the adverse effects of OPPs. MethodsThe lymphocytes of peripheral blood from 33 workers expo sed to dimethoate and 15 control people were isolated and treated with saline an d dimethoate respectively in vitro.RT-PCR technique was used to determine M 3 gene expression.Basal and inducible gene expression levels were measured. ResultsThere was no significant difference in basal gene expression level between ex posed group and control group,while the inducible gene expression level was sign ificantly higher in exposure group(1.92±1.07) than in control group(1.22±0.19) and basal level(1.49±0.45,P< 0.05) .No differences in basal and inducibl e gene expression level were found between male and female people in both expose d and control group.The level of inducible M 3 gene expression increased with t he increase in length of exposure time[<5 a:(1.69± 0.95) ,5~25 a:(1.91±1 .03),>25 a: (2.09 ±1.25),the latter was significantly different from that of <5 a(P< 0.05) ]. ConclusionAfter long-term expo sure to OPPs,the basal M 3 receptor gene expression level in the exposed worker s did not show any difference from the control group,but the inducible gene expr ession level(treated with dimethoate in vitro) was increased and related to the extent of exposure to dimethoate.
