摘要
目的 观察三氧化二砷 (As2 O3 )对人膀胱癌细胞株BIU 87的作用 ,并探讨其作用机制。方法 采用四氮唑蓝(MTT )法检测As2 O3 不同浓度、不同作用时间对BIU 87细胞株的生长抑制率 ;流式细胞术 (FCM )检测不同浓度As2 O3 作用 72h后细胞中凋亡相关蛋白Fas、bcl 2的表达及细胞周期变化。结果 As2 O3 可有效抑制BIU 87细胞株的生长 ,具有浓度、时间依赖性特点。Fas、bcl 2的表达与As2 O3 浓度增高分别呈正、负相关 (P <0 .0 5 ) ,且随As2 O3 浓度增高Fas、bcl 2的表达呈负相关 (P <0 .0 5 )。随As2 O3 浓度增高细胞周期被阻滞在G0 /G1期。结论 As2 O3 可有效抑制人膀胱癌BIU 87细胞株的生长增殖 。
Objective To investigate the effects of arsenic trioxide on human bladder cancer cell BIU-87 and study its mechanisms.Methods The BIU-87 cell was treated by various concentration of As 2O 3 for different period and the cell growth inhibition rate of BIU-87 cell was tested by MTT assay,Fas,bcl-2 expression and cell cycle were detected by flow cytometry after BIU-87 cell was treated with various conentration of As 2O 3 for 72 hours.Results As 2O 3 effectively inhibited the growth of BIU-87 cell and the inhibitory effect was related with the concentration of As 2O 3 and the period of treatment.The expression of Fas and bcl-2 was positively and negatively correlated with the increase of As 2O 3 concentration respectively(P<0.05).And with the increase of As 2O 3 concentration,Fas expression was negatively related with bcl-2 expression(P<0.05).BIU-87 cell cycle was arrested in G 0/G 1 phase by the increased As 2O 3 concentration.Conclusion As 2O 3 effectively inhibits the growth of bladder cancer cell BIU-87.Cell cycle arrest and inducing cell apoptosis maybe the mechanisms of the inhibitory effect.
