人转化生长因子-β1转基因表达对大肠癌细胞(HT-29)体内生物学行为的影响

摘要 为证实 TGF-β1转基因表达对 HT-29细胞株体内生物学行为的影响,将正反义 TGF-β1基因转染后克隆出的抗性细胞株分别接种棵鼠皮下。结果证实:TGF-β1基因表达对裸鼠移植瘤产生明显的抑制作用,导致出瘤时间延长,肿瘤增殖速度减慢。TGF-β1抑制大肠癌细胞发生发展作用机制可能与 TGF-β1基因表达产物启动细胞凋亡有关。 In order to demonstrate the influence on the biological behaviour of HT-29 cell line by the expression of transfected TGF-β_1 gene in it,the stable cell clones resistant to G418 from HT-29 transfected with sense and antisense TGF-β_1 gene were inoculated in the skin of the null mice(Balb/c,nu/nu).The results indicated obvious inhibitory effect of expression of TGF-β_1 gene on the growth of graft tumor in the skin of null mice.The time of tumor development was postponed,and the tumor grew slowly.The inhibitory mechanism of the tumor progression by TGF-β_1 may be related to apoptosis induced by the expression TGF-β_1 gene.
