摘要
目的 探讨发育期小鼠肝细胞核DNA倍体动态变化规律。方法 本文选取不同发育时期健康雄性小白鼠的肝细胞涂片 ,Feulgen染色 ,TIGER细胞图像分析仪测量单个完整肝细胞核的DNA含量与倍体 ,分析发育时期小鼠不同DNA倍体肝细胞核所占的比率。结果 不同鼠龄小鼠同一倍体的肝细胞核DNA含量大致相同 ;二、四、八、十六倍体的肝细胞核间的比值均接近 2、4、8,它们的CV <9% ;鼠龄 2 - 2 0天的小鼠以二倍体肝细胞核为主 ;35天小鼠具有四种不同DNA倍体的肝细胞核 ,各DNA倍体肝细胞核所占比例依次为 :4C >8C >2C >1 6C ;6 0天小鼠偶见三十二倍体肝细胞核。结论 小鼠肝细胞多倍体化出现的时间比已有实验结果更早。
Objective To find out the pattern of DNA ploidy changes of hepatocyte nuclei of growing mice after birth. Methods Smears of liver suspension from growing mice after birth were stained with Feulgen, DNA content of intact hepatocyte and the percentage of different DNA ploidy nuclei were analyzed by TIGER cell image analysis system.Results DNA content of hepatocyte nuclei with the same DNA ploidy is almost identical at different growing ages; IOD ratio among 2c, 4c, 8c and 16c is close to 2、4 and 8 respectivel, and the CV of IOD from four types of DNA content ploidy is less than 9 percent. Most of hepatocyte nuclei in 2~20 day old mice are diploid, the nuclei in 35 day old group contain four typical DNA ploidy, a descending order of frequencies in various DNA ploidy can be illustrated in the following manner: 4C>8C>2C>16C, the 32c nuclei occasionally appear in 60 day old mice. Conclusion Polyploidization of mice hepatocyte nuclei appear in earlier growing stage after birth and more intensity compared with previous study.
