三种亚型多巴胺受体mRNA在人神经干细胞中的体外诱导表达

摘要 目的 比较不同培养条件下原代神经球细胞中三种多巴胺受体亚型表达的变化 ,探讨发育过程中不同亚型多巴胺受体表达调控的机制。方法 流产胎儿脑组织经体外培养获得神经干细胞球 ,再用各种因子进行诱导分化 ,提取细胞总RNA并用RT -PCR方法检测mRNA转录。结果 在神经球细胞中三种多巴胺受体均有表达 ,中脑与海马来源的神经球细胞无显著差异。在无血清培养的条件下 ,表皮生长因子 (EGF) ,碱性成纤维细胞生长因子 (bFGF)及白介素 (Ils)诱导均使得D3 受体表达下调 ,而有血清培养时 ,白介素 (Ils)诱导则不能出现下调或关闭的结果。结论 因子诱导产生的分化对于D3 受体表达起下调或关闭作用。 Aim To compare the mRNA expression of three dopamine receptor subtypes(D 1,D 2,D 3) in neurospheres under different culturing conditions.Methods Neurospheres derived from aborted fetal brain were cultured in different media.Total RNA was extracted and the mRNA transcription was tested by RT-PCR.Results All three subtypes of dopamine receptors were expressed in neurospheres both from mesencephalon and from hippocampus.When cultured under no serum medium,ILs or EGF and BFGF induction caused the down regulation of the D 3 mRNA transcription.But in 10% serum-contained medium,D 3 mRNA expression did not changed.Conclusions The differentiation caused by induction maybe a critical mechanism to shut down D 3 mRNA transcription.Some unknown factors in serum may inhibit this process.
