HPLC-ELSD法测定芪地抗疲劳颗粒中黄芪甲苷的含量

Determination of Astragaloside Ⅳ in Qidikangpilao Granules by HPLC-ELSD

目的 建立HPLC ELSD测定芪地抗疲劳颗粒中黄芪甲苷含量的方法。方法 选用C18色谱柱 ,乙腈 -水 (1∶2 )为流动相 ,流速 1ml/min ,漂移管温度 4 0℃ ,氮气压力 3.4Bar下进行测定。结果 芪地抗疲劳颗粒中黄芪甲苷进样量在 0 .82～ 4 .10 μg与其色谱峰呈现良好线性关系 ,加样回收率为 97.72 %。结论 本方法简便、快速、准确 ,可作为芪地抗疲劳颗粒中黄芪甲苷的质量检测方法。

Aim To establish an HPLC-ELSD method for determination of astragalosideⅣ in Qidikangpilao granules.Methods The samples were analyzed on an ODS column,with mobile phase of acetonitrile/water (1∶2) at flow rate 1.0 ml/min.The temperature of drift tube for ELSD was 40℃ and the pressure of N 2 was 3.4 Bar.Results Linear correlations of astragalosideⅣ was found between peak areas and concentrations in the range of 0.82～4.10ug,and the recovery was 97.72%.Conclusion The method was easy,quick and accurate,and can be used for controlling the quality of astragalosideⅣ in Qidikangpilao granules.
