摘要
目的 探讨活血化淤中药对大鼠肝硬化制备过程中对肝线粒体的影响。方法 在大鼠肝硬化制备过程中经胃管灌服丹柴四君子汤 ,测定肝灌注液乳酸、肝组织匀浆MAO、SOD、MDA、ATP酶及进行肝细胞流式细胞学检查和光镜、电镜检查。结果 肝硬化组乳酸、MAO、MDA含量明显升高 ,SOD含量降低 ,肝细胞流式细胞学检查及肝纤维化程度 ,与中药组比较P <0 0 5。电镜肝细胞线粒体计数 ,肝硬化组与中药治疗组比较P <0 0 1。结论 活血化淤中药通过改善肝脏微循环的血流灌注及肝供氧 ,促进肝细胞新陈代谢 ,降低肝乳酸含量 ,维持细胞内钙稳定 ,保证电子传递链传递 ,提高F0 F1ATPase的活力 ,提高SOD活性 ,及时清除自由基 ,有效的保护肝细胞线粒体 ,从而减轻肝细胞损伤 ,抑制肝纤维化进程。
Objective To investigate the effects of a Chinese herbal compound with function of promoting blood circulation by removing blood stasis on hepatic mitochondria during infliction of liver cirrhosis in rats. Methods The rats were randomized into 3 groups. During infliction of liver cirrhosis, the rats in group A were fed with the Chinese herbal compound but those in group received no feeding. Then lactic acid (LC) content in the liver perfusate and MAO, SOD, MDA and ATPase contents in the liver tissue homogenate were determined. Meanwhile, hepatocytic flow cytometry, light microscopy and electron microscopy were performed. Results The contents of LC, MAO and MDA were significantly higher but that of SOD lower in group A than in group B. There were significant differences in analytical results of cytology and the degree of hepatic fibrosis between the 2 groups ( P<0.05). The number of mitochondria was markedly lower in group A than in group B ( P<0.01). Conclusions Through improving blood filling of hepatic microcirculation and supplying oxygen for the liver, the Chinese herbal compound can speed up hepatic metabolism, decrease LC content, maintain Ca 2+ stabilization in cells, assure respiratory chain, increase vigor of F 0-F 1 ATPase, improve activity of SOD and clean out free radical time. Therefore, the compound can relieve the liver cellular damage to restrain the course of hepatic fibrosis through effectively protecting hepatocytic mitochondria.
