摘要 目的 通过检测正常心肌组织及心肌梗死后干细胞因子 (SCF)mRNA的表达水平 ,为探讨其在心肌梗死中的作用奠定基础。方法 将成年SD大鼠随机分为正常组、结扎 6h组、结扎 1d组、结扎 3d组 (各时间点均设假结扎组以作对照 ) ,通过结扎左前降支建立心肌梗死模型 ,假结扎组只穿线不结扎。各组取心脏左室前壁同一部位 ,提取总RNA ,用RT PCR的方法 ,以GAPDH基因为内参 ,进行半定量检测正常心脏及梗死后SCFmRNA的表达水平。结果 可溶型SCF和膜型SCF在正常心肌有低水平表达。在心肌梗死后的三个不同时间点 ,结扎组心肌组织中可溶型SCF的表达分别为假结扎组的 1 6 6倍 (P <0 0 5 )、1 5 8倍和 1 30倍 (P <0 0 1) ;而膜型SCF的表达分别为假结扎组的1 80倍、1 5 3倍 (P <0 0 5 )和 1 5 7倍 (P <0 0 1)。结论 心肌梗死可以促使SCF的表达上调 ,提示其在心肌梗死后的病理过程中以及在诱导干细胞的归巢中可能发挥作用。 Objective To investigate the role of Stem Cell Factor (SCF) in heart infarction by detecting the mRNA expression of SCF in normal and infarct heart tissues. Methods Hearts from SD adult rats were used as models, which includeds: the normal heart group; the pseudo-ligation group and ligation group six hours, one day or three days after operations. Total RNA was extracted from the same place of the heart tissues. Reverse transcript polymerase chain reaction (RT-PCR) was used to examine the relative abundance of mRNA. Results The results indicated that the levels of expression for soluble SCF and trans-membrane SCF were low in the normal heart group. They were up-regulated at all three time points after ligation. The levels of soluble SCF were 1.66 fold (P<0.05), 1.58 fold and 1.30 fold (P<0.01), respectively compared with the pseudo-ligation group; while the levels of trans-membrane SCF were 1.80 fold, 1.53 fold (P<0.05) and 1.57 fold (P<0.01) of the pseudo-ligation group, respectively. Conclusion Heart infarction could up-regulate the expression of SCF. The increased SCF might contribute to the homing of stem cells, although further investigation is needed.
