基质金属蛋白酶-7基因及蛋白表达与软骨肉瘤的相关性研究 被引量:2

摘要 目的 基质金属蛋白酶在肿瘤侵袭和转移中发挥重要作用 ,本研究探讨MMP 7基因表达与软骨肉瘤发生的可能关系。方法 应用逆转录 聚合酶链反应 (RT PCR)和免疫组织化学(S P)方法 ,对 2 4例软骨肉瘤患者的肿瘤和瘤旁正常组织的MMP 7基因转录本和蛋白表达进行检测。结果 应用RT PCR方法检测 2 4例软骨肉瘤组织中MMP 7基因mRNA表达情况 ,结果显示MMP 7在软骨肉瘤和瘤周正常组织中的表达差异有非常显著性 (P <0 .0 1) ,MMP 7mRNA在软骨肉瘤组织中表达明显增高。软骨肉瘤中MMP 7蛋白表达阳性百分比为 66.7% (16/2 4) ,瘤周正常组织为 2 9.2 % (7/2 4) ,差异有非常显著性 (P <0 .0 1) ,MMP 7蛋白在软骨肉瘤中表达增高。结论 MMP Objective To study the possible correlation between the expression of MMP-7 and the carcinogenesis of condrosarcoma.Methods Matched normal and cancerous tissues from 24 patients with chondrosarcoma were analyzed by RT-PCR and immuneohistochemistry (SP) methods.Results There were significant difference in the expression of MMP-7 between chondrosarcoma and its normal tissue by RT-PCR (P< 0.01).In SP method,the positive rate of MMP-7 protein in 24 cases of chondrosarcoma was 66.7% (16/24),and 29.2% (7/24) in normal tissues (P<0.01).MMP-7 mRNA and protein were highly expressed in chondrosarcoma.Conclusion High expression of MMP-7 mRNA and protein in chondrosarcoma may play an important role in cancinogenesis and invasion of chondrosarcoma.
