^(99)Tc^m标记抗人粒细胞单抗SZ-102在荷人胰腺癌裸鼠的显像研究 被引量:1

摘要 目的研究^(99)Tc^m标记抗人粒细胞单克隆抗体SZ102在荷人胰腺癌裸鼠体内显像及生物分布。方法以2亚氨基噻吩盐酸盐(2IT)修饰SZ102,99Tcm葡庚糖酸钠(GH)配体交换法标记SZ102,经裸鼠尾静脉注入99TcmSZ102后4、8和24h分别测定荷瘤小鼠体内组织的放射性分布,并于1和4h对裸鼠进行γ显像。结果γ显像及生物分布示,99TcmSZ102静脉注入荷瘤鼠后1h,肿瘤清晰显影,随时间延长,瘤体内放射性越来越浓,且瘤组织与非瘤组织的放射性比值(TNT)也逐渐增高,各时相肿瘤每克组织百分注射剂量率(%IDg)均高于注射99TcmIgG的对照组。结论99TcmSZ102具有活体内定位导向能力,显像时间短。 Objective To study the image and biodistribution of ~99 Tcm labeled anti-granulocyte monoclonal antibody (McAb) SZ-102 in human pancreas carcinoma xenografts, and to develop new technology for early diagnosis of pancreas carcinoma. Methods The McAb SZ-102 was labeled with ~99 Tcm using 2-iminothiolane modification and ~99 Tcm-glu-coheptonate (GH) transchelation method. The distribution 4, 8 and 24 h after injection of ~99 Tcm-SZ-102 was observed, and the images were taken using γ scintigraphy at 1, 4 h postinjection. Results Clear tumor image was obtained 1 h after injection of ~99 Tcm-SZ-102. The radioactivity in the tumor and the radioactivity ratio of tumor to non-tumor tissue increased as time passing by. The %ID/g of tumor tissue in experiment group was higher than that in control group at different intervals. Conclusion ~99 Tcm-SZ-102 perhaps is a promising pancreas carcinoma imaging agent.
