摘要
目的探讨可降解纤维蛋白胶血管外周支持对静脉移植物内膜增生的影响。方法建立大鼠颈总动脉自体颈静脉移植模型,按照有无纤维蛋白胶血管外周支持,分为纤维蛋白胶血管外周支持组、无血管外周支持组,每组24只大鼠。术后1周、2周、4周分别切除移植静脉,计算机图像分析系统测量和计算内膜、中膜厚度,进行透射电镜检查,用免疫组织化学方法检测静脉移植物平滑肌细胞增殖细胞核抗原(PCNA)的表达。结果纤维蛋白胶血管外周支持组静脉移植物术后1~4周内膜、中膜的厚度,明显低于无血管外周支持组(P<0001)。纤维蛋白胶血管外周支持组静脉移植物的PCNA指数也明显低于无血管外周支持组(P<001~0001)。透射电镜检查显示纤维蛋白胶血管外周支持组静脉移植物破坏程度轻于无血管外周支持组。结论纤维蛋白胶血管外周支持可抑制血管内膜、中膜在移植术后的增生。
Objectives: To observe effect of fibrin glue perivenous support on neointimal thickening in the rat vein grafts Methods: Rat jugular vein grafting models made by transplanting autologous external jugular vein grafts to the carotid arteries in 48 rats, were divided into FS group (fibrin glue supported vein grafts) and NS group (without perivenous supported vein grafts) The vein grafts were harvested at 1, 2, 4 weeks after operation The thickness of neointima and media in the vein grafts was calculated by computer image analysis system Electron microscopic and PCNA immunohistochemistry were carried out Results: The thickness of neointima and media in the vein grafts in NS group surpassed FS group significantly ( p <0 001).The vascular smooth muscle cell proliferation in FS group was significently lower than that in NS group ( p <0 01~0 001) Electron microscopical examination in NS group showed an extensive destruction, but vein grafts had slight damage in NS group and almost were intact Conclusions: The fibrin glue perivenous support can reduce intimal and medial hyperplasia
