大鼠神经肌肉接点N型乙酰胆碱受体密度测定的ELISA法 被引量:1

摘要 目的研究大鼠神经肌肉接点乙酰胆碱受体密度测定的一种方法,并以之测定不同类型疲劳神经肌肉接点乙酰胆碱受体密度,以期揭示运动性疲劳在运动终极处的发生机理。方法用银环蛇毒包被96孔酶标板,以受体夹心法测定乙酰胆碱受体,探讨最适条件,并以之测定不同类型疲劳神经肌肉接点乙酰胆碱受体密度。结果a-银环蛇毒包被浓度为50mg/ml,乙酰胆碱受体粗提物滴度为1∶800,酶标蛇毒滴度为1∶2000,显色反应结束后OD值在0.2~0.3,可以明显区分一定范围内不同浓度的粗提物。乙酰胆碱受体密度(以吸光度表示)在疲劳发生后密度下降。结论该方法测定灵敏度在微克-纳克级,重现性良好,操作简单,能够满足实验要求。以此方法进行的测定结果表明,乙酰胆碱受体密度下降可能是肌肉疲劳在神经接点处的机理之一。
