摘要
通过正交实验用带有2~3片叶的单芽做外植体,探讨了培养基成分对非洲菊离体培养有效增殖的影响,结果表明:MS+BA1 0mg·L-1+NAA0 1mg·L-1+蔗糖40g·L-1是有效增殖的适宜培养基,其有效增殖系数达8~9,且丛生芽的生长势健壮,叶片叶绿素含量高。同时对增殖丛生芽中玻璃化苗的生理状况进行了分析,结果表明:玻璃化试管苗叶片的叶绿素a和叶绿素b含量以及干重/鲜重显著低于正常苗叶片,但叶绿素a/b值差异不显著。KT促进非洲菊增殖的活力比BA弱,增殖系数达3~4,但10mg·L-1的KT形成的丛生芽健壮,叶片舒展,叶绿素含量高。因此,可用BA与KT进行一定轮次的更换使用;活性炭的加入,极大地抑制了非洲菊试管苗增殖;增殖培养基中增加琼脂用量,可有效降低增殖苗的玻璃化程度,但增殖周期延长5~8d。
The effects of medium component on effective multiplication of gerbera buds were studied by orthogonal design using shoots with 2~3 leaves.The optimum medium for multipication of gerbera shoot was MS containing 10 mg·L-1 BA,01 mg·L-1 NAA,and 40 g·L-1 sucrose.The effective multiplication ratio was up to 8~9,shoot was vigorous,and contents of chlorophyll a and chlorophyll b was higher under the optimum medium.As the same time,the physiologic norms of vitreous multiplication shoot were analyzed,the results showed that DW/FW,content of chlorophyll a,chlorophyll b and total content of chlorophyll of vitreous leaf was obviously lower than normal leaf,but chlorophyll a/b was not obvious.Using in Gerbera multiplication,the vigour of KT was inferior than BA,multiplication ratio of KT was 3-4,but the shoot was more vigorous,the leaf extended,the content of chlorophyll was high.KT and BA taked several tums for Gerbera multiplication;AC restrained multiplication of Gerbera test tube plant;simultaneously increasing content of agar,the degree of vitrifiation of proliferation shoot can be decreased effectively,but the cycle of multiplication lengthened 5-8 days.
