摘要
目的 检测近视眼患者角膜上皮细胞单纯疱疹I型病毒 (HSV I) ,探讨其在正常角膜潜伏的可能性。方法 取行准分子激光屈光性角膜切削术 (PRK)的近视眼角膜上皮细胞 13 8份 (13 8眼 ) ,每份均匀分为三部分 ,两份分别行HSV I的原位聚合酶链反应 (ISPCR)检测和间接免疫荧光法 (IFA)检测 ,阳性者取其相对应的第 3份角膜上皮进行病毒分离培养和电镜观察。结果 13 8份近视眼角膜上皮中 ,ISPCR阳性者 8眼 (5 8% ) ,IFA阳性者 4眼 (2 9% ) ,两种检测均阳性者 3眼 (2 2 % ) ,取此 9眼的第 3份角膜上皮进行病毒分离培养 ,电镜扫描未发现病毒颗粒。结论 正常角膜上皮细胞内存有HSV I功能基因 ,提示可能有HSV
ObjectiveTo detect the HSV-I in corneal epi t helial cell with myopia and investigate the possibility of the HSV-I hiding in normal cornea.Methods138 corneal samples from 138 myopia eyes were obtained during PRK and each was equally divided into three parts,2 of 3 parts were scree ned by Indirect Immunofluorescence Assay (IFA) and in situ Polymerase Chain Reac tion (ISPCR) to evaluate the possibility of the HSV-I latency.Then the 3rd part of corneas with positive results was used for the isolation of HSV-I and elect ron microscope examination.ResultsEight (5 8%) of the 138 samples were posi tive for HSV-I DNA by ISPCR,4 (2 9%) by IFA,3 (2 2%) by both,and no HSV-I wa s found under electronic microscope.ConclusionHSV-I DNA is positive in norm al corneal epithelial cell,suggesting the possibility of HSV-I latency in norma l cornea.
