增生性玻璃体视网膜病变患者血清和眼内液HGF水平检测

Hepatocyte growth factor levels in vitreous,subretinal fluid and serum in patients with proliferative vitreoretinopathy

目的 检测视网膜脱离 (RD)和 /或伴有增生性玻璃体视网膜病变 (PVR)患者血清和玻璃体液、视网膜下液(SRF)中肝细胞生长因子 (HGF)的含量 ,并探讨其相互关系。方法 同时收集 3 5例RD患者血清和玻璃体液、SRF标本 ,酶联免疫吸附分析 (ELISA)方法检测HGF质量浓度 ,其中 9例不伴有PVR的单纯黄斑孔 (MH)患者作为对照组。结果 各组患者血清HGF质量浓度无明显差异 (P >0 0 5) ,明显低于同组眼内液的质量浓度 (P <0 0 1) ,且二者无相关性 (P >0 0 5)。PVR玻璃体内HGF质量浓度 (10 91± 3 87)ng/ml,均比对照组 (3 42± 0 85)ng/ml高 (P <0 0 1) ,但PVRC级和D级之间差异不明显 :(7 88± 3 83 )ng/ml,(10 2 4± 4 0 7)ng/ml,P >0 0 5。SRF外引流 (14 68± 1 16)ng/ml或内引流 (10 3 5± 6 94)ng/ml的HGF质量浓度均高于玻璃体内水平 (P <0 0 5)。结论 PVR眼内HGF高质量浓度不是通过血 -视网膜屏障弥散 ,可能来自眼组织细胞的分泌 ;玻璃体内HGF随着PVR的发展而增高 。

ObjectiveTo study the relationship between hepatocyte gr ow th factor (HGF) in intraocular fluid and proliferative vitreoretinopathy (PVR). MethodsSerum,vitreous fluid and subretinal fluid (SRF) were simultaneously c ollected from 35 patients with PVR and 9 patients with macular hole (control).Co ncentrations of HGF were determined by enzyme-linked immunosorbent assay (ELISA ).ResultsThe mean HGF concentrations in serum had no significant difference in various groups and were lower than that in intraocular fluid ( P <0 01),b ut there was no correlation between two samples.Intravitreous level of HGF was h igher in PVR patients (10 91±3 87 ng/ml) in comparison with control patient s (3 42±0 85 ng/ml)( P <0 01),but there was no statistical differences between PVR-C degree and D degree (7 88±3 83,10 24±4 07 ng/ml)( P >0 05).SRF HGF concentrations were higher in RRD (14 68±1 16 ng/ml) and PVR (10 35±6 94 ng/ml) than that of vitreous fluid in PVR ( P <0 05).Conc lusionThe intraocular HGF in PVR patients may be derived from the secretory fu nctions of ocular tissues.With the development of PVR,vitreous HGF levels increa se,suggesting that HGF involves in the pathological process of PVR.
