c-myc、bcl-2基因表达和蝌蚪提取液抗早幼粒细胞白血病作用的研究

摘要 目的 研究蝌蚪提取液 (T871)抗早幼粒细胞白血病 (APL)的作用及其过程中c myc、bcl 2癌基因表达的变化。方法 利用细胞生长曲线、形态学观察、流式细胞仪、TUNEL法及原位mRNA杂交和完整细胞原位斑点印迹技术 ,观察T871抗人APL细胞系 (HL 6 0 )细胞的作用及其过程中c myc、bcl 2癌基因表达的变化。结果 T871能抑制HL 6 0细胞增殖 ,并出现细胞凋亡的形态学特征 ,流式细胞仪及TUNEL法检测结果显示凋亡细胞百分数比对照组显著增加 ,并伴有癌基因c myc、bcl 2mRNA表达的下调。 结论 T871可抑制HL 6 0细胞的增殖同时诱导其凋亡 ,且c myc、bcl Objective To investigate the function of antileukemia by tadpole extract(T871)and expression of c-myc,bcl-2 in the process.Methods We studied the effects of T871 on proliferation and apoptosis of human promyelocytic leukemia cell line(HL-60)cells by cell growth stage,cytomorphological observation,flow cytometry analysis and TUNEL method.We also examined the oncogene(c-myc?bcl-2)expression during the induction of apoptosis in T871 treated HL-60 cells by in situ hybridization and intact cell mRNA dot blot techniques.Results T871 inhibited the proliferation of HL-60 cells.T871 induced HL-60 cells to apoptosis as evidenced by:(1)characteristic morphology of apoptosis.(2)The flow cytometry analysis and TUNEL methods analysis showed an increase in the number of apototic HL-60 cells increased.The apoptosis of HL-60 cells was accompanied by down-regulation of c-myc,bcl-2 oncogene expression.Conclusion Tadpole extract may induce HL-60 cells to apoptosis and oncogene c-myc,bcl-2 may involve in the apoptotic course.
