彩色多普勒超声对胎鼠心肌细胞凋亡影响的实验研究 被引量:5

摘要 目的 探讨诊断用彩色多普勒超声是否对胎鼠心肌细胞构成影响。方法  2 4只孕 14天SD大鼠按彩超辐照时间不同随机等分为四组 ,Ⅰ组 (对照组 )、Ⅱ组 (照射 10min组 )、Ⅲ组 (照射 2 0min组 )、Ⅳ组 (照射 3 0min组 ) ,照射条件 :4V1探头 ,二维频率H3 .0MHz ,彩色频率CD =3 .0MHz ,软组织热指数TIS =1.8,机械指数MIcd =1.6。照射后 2 4h剖宫取胎鼠心肌标本 ,HE染色光镜观察 ,TUNEL法检测凋亡细胞。结果 各组光镜下观察均未见异常 ,TUNEL法检测结果 ,对照组(Ⅰ )及照射 10min组 (Ⅱ )、照射 2 0min组 (Ⅲ )胎鼠心肌组织内仅可偶见散在凋亡细胞 ,荧光强度各组间无显著差异 (P >0 .0 5 ) ;照射 3 0min组 (Ⅳ )凋亡细胞较易观察到 ,部分区域可见较密集的凋亡细胞。照射 3 0min组荧光强度与对照组 (Ⅰ )有显著差异 (P <0 .0 5 )。结论 诊断用彩色多普勒超声照射孕鼠 3 0min以上有可能引起胎鼠心肌细胞凋亡。 Objective To study whether diagnostic CDUS can induce fetal rat's myocardial cell apoptosis. Methods Twenty-four female Spraque-Danles rats that had been pregnant for 14 days were divided into four groups averagely and randomly, control group (groupⅠ) was sham-irradiation, groupⅡ-Ⅳ were irradiated by diagnostic CDUS (Sequoia-512, 4V1, H 3.0 MHz, CD=3.0MHz, TIS=1.8,MIcd=1.6) for 10 minutes, 20 minutes and 30 minutes respectively, fetal rats' hearts of four groups were removed in 24 hours after irradiation, HE Stain was done for microscope observing, apoptosis cells were detected with TUNEL (in situ terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling). Results There were no abnormal changes among groupⅠ-Ⅳ under microscope, and there was no significant difference in apoptosis among groupⅠ, Ⅱand Ⅲ ( P >0.05), but apoptosis cells increased in group Ⅳ. Conclusion Continuous irradiation beyond 30 minutes by diagnostic CDUS may result in fetal rat's myocardial cell apoptosis.
