抗伊氏锥虫单克隆抗体的研究——二 、用McAb直接斑点法检测伊氏锥虫抗原的实验研究

摘要 抗伊氏锥虫糖蛋白的单克隆抗体ⅡG_6标记过氧化物酶后,用于直接法斑点试验检测血清中伊氏锥虫(T.e.)抗原,36份感染2—4d的鼠血清全部呈阳性反应;12份正常鼠血清全部呈阴性反应.上述结果表明本方法具有较好的敏感性和特异性.ⅡGMcAb Dot ELISA检测结果显示血中伊氏锥虫密度与抗原滴度呈正相关.该法操作简便易行,不需特殊设备.肉眼即可观察结果.本次实验研究将为现场应用提供快速诊断的重要依据. The monoclonal antibody IIG6 (against ghycoprotein of Trypanosoma evansi ) labelled with peroxidase was used in Dot-ELISA test for detecting trypanosoma antigen in serum. Positive reactions were found in specimens of serum from 36 mice infected in 2-4 days with Trypanosoma evansi and negative in 12 normal mice. The above results showed that the sensitivity and specificity of direct Dot-ELISA with monoclonal antibody were high enongh for detecting the trypanosoma antigen. A positive correlationship between antigen tisters and circulating Trypanosoma evansi density was observed with IIG6 McAb Dot-ELISA. Dot-ELISA has a certain advantage over ELISA for its simpler procedure and no special equipment such as photometer in needed. This technique is suitable for field work and convenient for epidemi-ological study.
