脐血细胞的生物学特性的研究

Research on biological characteristic of cord blood cell

目的 探讨脐血造血干/祖细胞的质和量以及淋巴细胞的表型特征。方法 收集34份脐血,利用6％的羟乙基淀粉沉淀红细胞,用甲基纤维素半固体培养法观察造血干/祖细胞的增殖能力,流式细胞仪检测CD34^+细胞的数量和淋巴细胞的表型。结果 34份脐血的有核细胞回收率为79.2％±13.7％,平均有核细胞数为(12.2±4.3)×10~8;BFU-E、CFU-GM分别为(2.05±1.22)×10~6、(3.14±1.43)×10~6;CD34^+细胞数平均为(1.98±1.30)×10~7;CD4^+T淋巴细胞表达CD45RA达90.97％±6.96％,CD8^+T淋巴细胞表达CD45RA达98.93％±0.36％,均显著高于正常成人外周血淋巴细胞(P<0.01)。结论 单份脐血的干/祖细胞可以满足成人移植的需要,且脐血中的淋巴细胞功能不成熟,同种异体的免疫反应弱,用于移植时移植物抗宿主病(GVHD)的发生率可能较低。

Objective To elucidate the quality and number of hcmatopoietic stem/progenitor cells as well as im-munophenotypic features of lymphocyte in cord blood. Methods Analysis of the capacity of colony forming unit was per formed by means of methylcellulose culture method and immunophenotype was analysed by FACS using a large panel of McAb in double labelling in 34 cord blood samples. Results Of 34 cord blood samples, recovery of nucleated cell is 79.2% 13. 7% ,average number of nucleated cell is (12. 2X.3)X108.In methylcellulose medium,average colony yield of BFU-E and CFU-GM is (2. 051. 22) X 106 and (3. 141. 43) X 106 respectively. The number of CD34 is (1. 981. 30)X 107. The large majority of HUGE T-cells co-expressed CD45RA native antigen.CD4 CD45RA is 90. 97% 6. 96%.CD8 CD45RA ' is 98. 93% 0. 36%. Significant difference in precentagc were found between cord and adult T-cells ( P <CO. 01). Conclusion Single cord blood can meet the need of transplantation of one adult. The immunologic function of lymphocyte in cord blood is not mature ,immunoreactive potential to homogeneity is weaker.so the incidence and severity of Graft-Ver-sus-Host-Disease(GVHD) is lower after transplantation.
