献血人群HCV携带者NS5b区基因多态性的初步探讨

Preliminary discussion of NS5b region polymorphism of HCV carriers from blood donors

目的 探讨献血人群中丙型肝炎病毒(HCV)携带者NS5b区基因多态性。方法 对广西、安徽、西安、江苏、上海五个地区184例抗-HCV阳性献血员的血清用逆转录聚合酶链反应(RT-PCR)进行了HCVRNA检测,并对HCVRNA阳性血清用限制性片段长度多态性方法(RFLP)分析。结果 共检出42例HCVRNA阳性血清,其中广西8例,安徽4例,西安2例,江苏9例,上海19例。对这42份血清进行RFLP分析。结果显示:HCV 1b型37例(37/42,88.1％),HCV2a型5例(5/42,11.9％)。其中1b型中广西7例,安徽4例,江苏9例,上海17例;2a型中广西1例,上海2例,西安2例。进一步的实验表明,42例PCR产物全部都具有Hae Ⅲ酶切点,且5例2a型表现出3种基因变异。结论 我国献血人群中HCV感染者大多数属1b型,且HCV携带者的HCV基因在亚型下存在多种有规律的变异。

Objective To analyse the gene polymorphism of HCV NS5b region. Method The RT-PCR was used to screen 184 serum samples from Guangxi、 Anhui、Jiangsu、Xi' an and Shanghai, which are HCV antibody positive. On those RT-PCR positive products, RFLP analyses have been taken. Results 42 sera were RT-PCR positive. Among them,37 are HCV 1b type (88. 1 % ) . 5 are HCV 2a type (11. 9%). Within those 37 HCV Ib type samples,7 are from Guangxi,4 are from Anhui ,9 are from Jiangsu and 17 are from Shanghai. For HCV 2a type samples, 1 is from Guangxi,2 are from Xi' an and 2 are from Shanghai. The results showed that samples from these four areas are mainly HCV Ib infection. In addition,the NS5b fragments of the 42 HCVRNA positive are all have Haelll restriction sites. And there are 3 kinds of gene variations in 2a types. Conclusion There may be some subtypic gene variations in domestic HCVRNA positive samples. Those variations can be used to make HCV classification、epidemiological survey and tracing of HCV infection.
