摘要
目的建立测定家兔血浆中桂竹糖芥强心甙G的高效液相色谱法。方法选用Shim-packCLC-ODS15cm×6.0mmC185U色谱柱,流动相为乙腈-水-甲醇(2.5:60:15),以咖啡因为内标物,血浆样品经甲醇提取,N2吹干,流动相定溶,20μl进样,二极管阵列检测波长218um。结果线性范围为36.2-36200ugL-1,最低检测浓度为36.2ugL-1,精密度、准确度亦符合有关要求。结论该分析方法简单、灵敏、专属性强。
Objective A high Performance liquid chromatographic method for the quantitative determination of Erysmoside G, a new kind of cardiac glycosides, in rabbit plasma is presented.Methods Using a Shim-pack clc-ODS C18 column and a mobile phase containing acetonitrile:methanol:water (25: 65:15), with caffeine as the internal standard' The plasma samples have been extracted with methanol (1: 5), dried with N, at low temperature then dissolved and fixed at 1ml with mobile. 20ul of the solution has been injected into the chromatographic system. The analysis is detected by SPD - M10A at 218nm.Results Calibration curves are linear over the range of 36.2-36200ug/L.The detection lindt is 36.2 ug/L for Erysimoside G in the plasma.Conclusion The method proves to be simple and sensitive with good specificity, precision and accuracy.
