脑出血对大鼠不同部位脑组织中Ref-1表达的影响

摘要 目的 观察正常大鼠脑组织中氧化还原因子1(Ref-1)的表达状况;探讨脑出血对大鼠某一时间点(48 h)不同部位脑组织中Ref-1表达的影响规律。方法 用立体定向技术制备大鼠脑出血模型,将动物随机分为正常对照组、假手术对照组和出血模型组(ICH)。根据实验要求的时间点断头取脑,并制成石蜡脑片,连续切片做HE染色和(或)Ref-1免疫组化染色。结果 ①脑细胞内可见点状或片状Ref-1表达阳性的棕黄色颗粒存在,表达的部位主要在细胞核。②假手术对照组不同部位脑组织中,海马区Ref-1表达最显著(P<0.01)。③与假手术对照组的相同部位脑组织中Ref-1表达状况比较,脑出血对右侧尾状核区(血肿周围)脑组织的影响最显著(P<0 01),其次是血肿侧海马区(P<0.05)。结论 大鼠脑组织Ref-1阳性表达主要是在神经元细胞核内;Ref-1表达在正常对照组和假手术对照组脑组织的不同部位表达是相异的,相对海马区表达更明显;脑出血对大鼠某一时间点(48 h)不同部位脑组织中Ref-1表达下降程度的影响是不相同,与血肿远近有关。 Objective To investigate the expression of Ref-1 in rat's brain tissues at different regions in dummy group as well as the regular pattern in expression of Ref-1 in rat's brain tissues at different regions in the given time slice(48h) after intracerebral hemorrhage. Methods ICH was introduced in rat by using stereotactic infusion autologous blood 50μl into the caudate nucleus. The male animals were randomly divided into 3 groups: controlled group,dummy operation group and hemorrhage group. At the due time the rats were killed for brain tissues by means of paraffin section and immunohistochemistry method was employed to detect the expression of Ref-1 in cerebral tissues. Results In the dummy operation group, compared with the different regions ( round clock of 48h), the expression Ref-1 positive cells in the hippocampus was higher than that at other region in the brain tissue (P<0.01). In hemorrhage group,compared with the dummy operation group in the same region in the brain tissues near the caudate nucleus,the expression Ref-1 positive cells markedly decreased (P<0.001). Conclusion The expression Ref-1 positive cells in the controlled group mainly present in nucleus of neuron cells in the brain tissues; in the dummy operation group,the degree of Ref-1 expression Shows difference in distinct cerebral regions and significantly higher in the hippocampus; in the given time slice (48h) after ICH,Ref-1 expression is different in distinct regions and related to the distance from hematoma.
