人表皮生长因子(hEGF)及其受体(hEGF-R)通过^(125)Ⅰ标记分析法在21例结肠癌、11例正常对照标本中全部表达。hEGF在癌组织、癌周组织、癌远侧组织中表达无明显差异,但均高于正常对照组(P<0.05)。而hEGF-R在上述4种组织中逐次下降,差异显著(P<0.01)。hEGF-R在癌组织中表达随肿瘤分期而变化(P<0.01),与结肠癌的周围淋巴结转移相关(P<0.05),但与癌组织类型无关。同时,癌组织中hEGF-R的组织亲和力(kd)也随肠癌的分期而变化(P<0.01)。本文表明,hEGF及hEGF-R在结肠癌的发生中可能起癌基因的作用,并且hEGF-R随癌的进展、转移而其表达不断增强,从而有可能为结肠癌的中、早期诊断和治疗提供新的探索途径。
The expression of human epidermal growth factor (hEGF) and hEGF receptor (hEGF-R) was examined by 125I-labeled-hEGF binding analysis in 21 cases of colonic carcinomas. No significant difference was found in the values of hEGF binding among carcinoma, paracarcino-ma regions and regions far away from carcinoma, but the values in all of them were higher than that in normal colonic tissues (P<0.05). hEGF-R expression among the four groups were significantly different (P<0.01). hEGF-R expression varied with the stage of carcinoma (P<0.01). hEGF-R expression was correlated with carcinoma lymphatic metastasis (P<0.05), while it was not correlated with histological differentiation. The affinity of hEGF-R in cancer tissues varied with the stage of carcinoma (P<0.01). Our findings suggesed that hEGF-R may play a role of on-cogenesis. All of these give evidence for early diagnosis and treatment of colonic carcinomas.
