摘要
目的 : 预测和表达人角蛋白 17HLADR4、7限制性T细胞抗原表位区。方法 : 根据相关计算机软件和互联网服务器进行表位预测。从银屑病皮损表皮中提取总RNA ,反转录成cDNA ,常规方法将目的基因插入原核表达载体pGEX - 4T - 1和真核表达载体pCR3.1,表达和纯化相应蛋白和表位肽段。结果 : (1)预测得到了 5个人角蛋白 17HLADR4、7限制性T细胞抗原表位区 ;(2 )表达得到了人角蛋白 17及其HLADR4、7限制性T细胞表位肽段。结论 : 人角蛋白 17及其HLADR4。
Objective: To predict and express the HLA DR4,7-restricted T cell epitope regions on human keratin 17. Methods: Epitope prediction was based on related software and Internet servers. Total RNA was isolated from epidermis of psoriasis patients and mRNA was reversely transcribed into cDNA. The epitope genes were inserted into prokaryote (pGEX-4T-1) and eukaryote expression vector (pCR3.1). Keratin 17 and its epitope regions were expressed and purified.Results: (1)Five predicted HLA DR4,7-restricted T cell epitope regions were obtained. (2) Human keratin 17 and it's HLA DR4,7-restricted T cell epitope regions were expressed. Conclusion:The successful expression and purification of keratin 17 and its HLA DR4,7-restricted T cell epitopes may play a remarkable role in understanding the immunological pathogenesis and provide a basis for immunotherapy of psoriasis.
