高糖合并温热下小鼠巨噬细胞对BGC-823人胃癌细胞的杀伤作用

摘要 应用自然杀伤试验-LDH释放改良法测定昆明种小鼠腹腔巨噬细胞对BGC-823人胃癌细胞的自然杀伤作用.结果显示,在巨噬细胞与胃癌细胞的混合培养液中加入不同浓度的糖(160mg/dl和300mg/dl)增加了巨噬细胞对胃癌细胞的杀伤作用(P<0.05);同时,实验表明,在温热条件下(39℃~41℃)未见巨噬细胞对胃癌细胞的杀伤作用随培养温度的升高而提高,因而认为主要是因高糖条件下提高了肿瘤细胞的热敏感性和降低了肿瘤细胞的pH,从而促进了巨噬细胞对肿瘤细胞的杀伤作用. In this study,natural killing test-improved LDH release method was used to determine the natural killing effect of macrophages in mouse peritoneal cavity on human stomach cancer cell line BGC-823.The results showed that different concentrations of glucose (160mg/dl and 300mg/dl) added to the combined medim of macrophages and stomach cancer cells significantly increased the killing effect of macrophages on stomach cancer cells (P<0.05),but that when heated (39℃~41℃),the killing effect of macrophages on stomach cancer cells was not enhanced with the elevation of culture temperature Therefore,we conclude that hyperglycistia can increase thermal sensitivity and decrease the pH of tumor cells,thus improving the killing effect of macrophages on tumor cells.
