摘要
探讨成骨细胞在金黄色葡萄球菌感染后细胞因子的产生情况。方法 半对数生长期金葡菌6 5 71为实验用菌株。成骨细胞培养自健康成人下颌第三磨牙拔除术中新鲜去骨块。实验组以MOI为 30的比例感染成骨细胞 2小时 ;对照组未加细菌。在 0 ,2 ,4,8,12 ,2 4,48,72小时收集培养液 ,用ELISA方法测定样本中TNF -α、IL - 1β、IL - 6、IL - 8浓度。 结果 实验组和对照组均未见TNF -α、IL - 1β表达。实验组各时间点IL -6、IL - 8浓度明显高于对照组。结论 成骨细胞在受金葡菌感染后TNF -α、IL - 1β、IL - 6、IL - 8表达上的变化可能有利于机体对骨组织金葡菌感染的清除。成骨细胞可能是减轻金葡菌骨髓炎的重要细胞成份。
Objective Cytokine expressions in Staphylococcus aureus-infected human osteoblasts were studied.Methods Mid-logarithmic growing Staphylococcus aureus 6571 was prepared.Human ostoblasts were cultured from bone explants.Osteoblasts of experimental group were infected by Staphylococcus aureus at MOI 30 for 2 hours.Control group received the same treatment as the experimental group except adding of Staphylococcus aureus.Supernatants were collected at different time points.TNF-α、IL-1β、IL-6 and IL-8 proteins were assayed by ELISA.Results Neither of the two groups had TNF-α and IL-1β detected.Osteoblasts of the experimental group produced much more IL-6 and IL-8 than those of the control group.Conclusion Changes of TNF-α、IL-1β、IL-6 and IL-8 production in osteoblasts after being infected by Staphylococcus aureus may benefit the removal of Staphylococcus aureus from bone tissue,and osteoblasts are important cells to fight against osteomyelitis caused by Staphylococcus aureus.
