摘要
1目的 从形态学角度观察兔骨髓间充质干细胞体外培养的生长特性 ,建立一套体外分离、培养骨髓间充质干细胞的可靠方法。 2方法 取兔骨髓后 ,通过离心 ,贴壁培养 ,分离出间充质干细胞。在倒置显微镜下连续观察细胞的形态变化并记录其特点。 3结果 随时间的推移可见细胞由贴壁前的圆形变成椭圆形、梭形 ,少部分呈多角形 ,细胞呈漩涡样生长。同时可见脂肪细胞出现 ,部分细胞由梭形变为圆形 ,周围可见光强反射基质形成 ,类似软骨细胞。然后细胞增殖由快变慢 ,更换特制培养液细胞增殖速度加快。 4结论 兔骨髓间充质干细胞在体外可向成纤维细胞、脂肪细胞、软骨细胞和成骨细胞转化。
Objective To observe the growth character of rabbit's bone marrow derived mesenchymal stem cells (MSCs) morthologically in vitro, finding a proper set of culture system to isolate and cultivate MSCs in vitro.Methods Bone marrow was aspirate from rabbit, and MSCs were isolated by centrifugation and cultured in monolayers. Observed the cells through inerted microscope constantly and took records.Results The MSCs originally appeared in cluster of large stellate cells growing outward in a swirling pattern. During the growth, some of the adherent cells progressed from spindyloid,fibroblast-appearing cells to rounder cell with refractile matrix, resembling chondrocytes, some differentiated into adipocytes, most of them remained spindyloid.Conclusion MSCs have the capacity to differentiate into fibroblast,adipocyte , chondrocyte and osteoblast. Cell growing factors can strengthen the capacity. [
