不同摇瓶条件对侧耳菌生长及漆酶分泌的影响 被引量:6

摘要 研究了小试液体摇瓶下不同pH培养条件、摇瓶转速、装液量对侧耳属白腐菌BP漆酶分泌及生长状况、形态变化的影响。结果表明 :BP在中、酸性条件下生长较好且表现较高的分泌漆酶能力 ,用硫酸调pH为 5 .0时 ,漆酶活力和生物量分别在第10d和第 6d达到最高 ,分别为 74 2U mL和 8.4g(干重 ) L ,高于相应缓冲液调节pH体系的结果 ;天然 (pH6 .5 )培养条件下 ,最佳转速与装液量分别是 15 0r min、2 0 0mL 5 0 0mL三角瓶 ,此时菌球较小 ,四周为毛刺状 ,漆酶酶活和生物量分别在第 5d和第 6d达到最高 ,分别为 714 .2U ml和 9.2g L。同时 ,运用SPSS统计软件对各影响因素进行差异显著性检验 ,获得白腐菌生长及漆酶分泌特性的主要影响因子。 The effects of different pH, shake flask agiation speed and culture volume conditions on pleurotus white rot fungus BP's producing laccase,growth and morphological changes have been studied.The results showed that more biomass and higher laccase activity can be obtained in neutral or acid conditions.The most laccase activity and biomass are both achieved in pH5.0 medium,but which are more in buffering system than in non-buffering system and are about 742U/mL and 8.4g/L in 10^(th) and 6^(th) day respectively.The relative optimum speed and culture volume is 150r/min and 200mL/500mL respectively in natural mediums(pH6.5).At this time,pellets are small and rough surface, the most laccse activity and biomass can achieve 714.2U/ml and 9.2g/L in 5^(th) and 6^(th) day respectively.The SPSS statistical software was applied to check those remarkable differences for varies factors.
