摘要
采用不同浓度的秋水仙碱对绿玉树试管丛芽块进行多倍体诱变处理 ,试验结果表明 :不同处理方法和处理浓度影响四倍体的诱变率 ,0 .2 5 %秋水仙碱浸泡和共培养 4 8h以上均能诱导绿玉树产生大量的四倍体小苗。四倍体小苗与正常二倍体小苗比较 ,叶子颜色深绿 ,不易脱落 ,茎干较粗壮 ,经根尖细胞学鉴定染色体鉴定为 4 0条 (2n =4x=4 0 ,x =10 ) ,耐低温能力强。
Experiments of polyploid inducement using colchicines was conducted on the Euphorbia tirucalli(L.). The results showed: the tetraploid proliferation rate was affected by concentration of colchicines and treatment methods. Explants whether were soaked in 0.25?% colchicine solution or cultured on media containing 0.25?% colchicines could proliferate tetraploid shoots. Compared with the shoots reproduced from control explants, polyploid shoots have black green leaves, larger stems, and leaves remained longer. The normal plantlet has 20 chromosomes, and the polyploid plantlet has 40 chromosomes. The tetraploid plantlets has greater cold-resistance.
