摘要
采用二次丙酮法与 75 %硫酸铵沉淀法从蘑菇 (Agaricusbisporus)中提取多酚氧化酶 ;用分光光度法对蘑菇中多酚氧化酶酶学特性进行了较为系统的研究。结果表明 :二次丙酮法优于 75 %硫酸铵沉淀法 ;以邻苯二酚为底物 ,多酚氧化酶的最适pH为 7.0 ,最适温度为 30℃ ;亚硫酸钠、半胱氨酸、抗坏血酸为强抑制剂。
The two-step acetone and 75 percent ammonium sulfate precipitation methods were used to extract polyphenoloxidase from agaricus bisporus. With spectrophotometry method, a systematic research on the characteristics of polyphenoloxidase in agaricus bisporus was made.The results indicted that the two-step acetone was better than the other one. Catechol as the substrate,the optimum pH and temperature were 7.0 and 30℃,sodium sulfite,cysteine, ascorbic acid showed a strong inhibitory effect.
