摘要
从20株抗支原体MAb中筛选出1株能够对各种支原体起反应的MAb(3D1)组成试剂盒,用间接免疫荧光法(IFA) 检查了35株传代细胞和6份动物标本,支原体检出率为83%,为了验证此方法的准确性,将其中部分细胞同时做常规培养检查,符合率为90%。结果证明,MAb试剂盒具有特异性高,敏感性强,速度快等优点。
A hybridoma(3D1) to secrete monoclonal antibody against most mycoplasmascausing cell culture contamination was chosen from 20 hybridomas for assembling a diagnostic kit. Using the diagnostic kit by IFA, 35 samples of continuous cell lines and 6 animal specimens wereexamined. Mycoplasma positive rate was 83%. In order to confirm the specificity and sensitivity ofthe kit to mycoplasmas, monoclonal antibody IFA was compared with routine mycoplasma cul-ture. All positive samples detected by routine mycoplasma culture were positive for monoclonalantibody IFA and a few positive samples detected by monoclonal antibody could not be detectedby routine mycoplasma culture. The results showed that monoclonal antibody IFA was more sen-sitive than routine mycoplasma culture. The monoclonal antibody diagnostic kit provided a rapid,simple and sensitive method for detecting mycoplasmas.
