分泌抗人IgA单克隆抗体杂交瘤细胞株的建立与鉴定

Establishment and characterization of the hybridoma producing monoclonal antibody against human IgA

利用常规杂交瘤技术制备了一株抗人ＩｇＡ杂交瘤ＰＡ２２９。ＥＬＩＳＡ结果表明，它分泌的抗体与人ＩｇＡ有特异性反应，而与其它免疫球蛋白重、轻链无交叉反应，为小鼠ＩｇＧ１亚类。免疫转印结果只显示针对ＩｇＡα链的一条电泳带。用改良ＥＬＩＳＡ法测得ＰＡ２２９的亲和常数为１．３３×１０＾８Ｌ／ｍｏｌ。

One cell line stably producing monoclonal antibody (McAb) was raised using rou- tine hybridoma technique and named as PA_(229). This McAb of mouse IgGl subclass reacted only with human IgA but did not react with the heavy and light chains of other human immunoglobulins in enzyme-linked immunosorbent assay (ELISA). The immunoblotting result showed that PA_(229) McAb recognized one band of 2 chain of human IgA. The affinity constant of PA_(229) McAb was measured to be 1.33×10~8 L/mol by an adapted ELISA. This McAb with a high degree of specificity to human IgA will be useful for nasopharyngeal carcinoma diagnosis.
