摘要
通过PCR和体外重组技术对TGFβ_1基因5′和3′非翻译区进行了改造,在此基础上,通过构建缺失突变体,采用双脱氧终止法,利用pUC/M13系统测定了人TGFβ_1cDNA的全长序列。这为今后研究不同长度非翻译区对TGFβ_1cDNA表达水平的影响并实施其高效表达奠定了基础。
The untranslated region of human transforming growth factor-β_1(TGFβ_1) gene
was modified via polymerase chain reaction and DNA recombination. After Sanger's dideoxy
chain-termination method, the sequence data of TGFβ_1 cDNA was obtained from pUC/M13
system by the construction of deletion mutants. This work will benefit the high level expression of
TGFβ_1 cDNA in the future.
