摘要 为了提高磷脂产品中磷脂酰胆碱 (PC)的含量 ,研究了以浓缩大豆磷脂为原料 ,采用丙酮和乙醇作溶剂 ,制备高含量磷脂酰胆碱。用丙酮脱油时 ,在料溶比为 15∶2 0 0、提取时间 15min、提取温度 4 0℃的最佳脱油工艺条件下 ,得到丙酮不溶物含量为 97.6 %的中间产物———粉末磷脂。用乙醇提取富集其中的磷脂酰胆碱 ,通过单因子试验 ,研究了粉末磷脂与溶剂的比例、乙醇浓度、提取温度、时间和次数对PC含量和提取率的影响。试验结果表明 ,在乙醇浓度 95 % ,固液比 1∶2 0 ,提取时间 30min ,温度 35℃ ,抽提次数 2次的条件下 ,PC含量和提取率均较优。在最佳试验条件下 ,PC含量可提高到 4 5 .1% ,提取率为 6 2 .0 %。 In order to improve the content of phosphatidylcholine(PC),a study has been carried out on the use of acetone and alcohol to extract PC from soybean concentrated phosphatide.The results showed that acetone insoluble residue of phosphatide powder was up to 97.6% by acetone extracting soybean concentrated phosphatide with the conditions of the ratio of acetone to soybean concentrated phosphatide 200∶15,extraction time 15 min and extraction temperature 40℃.The obtained phosphatide powder was further fractionated with ethanol.The effects of alcohol concentration,phosphatide powder/ alcohol,extraction time,temperature and times on the yield and PC content were evaluated.The yield and PC content were better under the conditions with the alcohol concentration 95%,phosphatide powder/ alcohol 1∶20,extraction time 30 min,temperature 35℃,extraction twice.Under the optimum conditions,the PC content and yield were 45.1% and 62.0%,respectively.
