摘要 目的 研究 β1转化生长因子 (TGF β1)对瘢痕成纤维细胞纤维粘连蛋白 (FN)及其受体α5β1整合素表达的影响 ,探讨TGF β1、FN及其受体α5β1整合素在增生性瘢痕形成发展中的作用。方法 采用细胞培养、ELISA法、免疫组织化学、图像分析技术 ,观察浓度分别为 0、5、10、2 5、5 0、10 0 μg/L的TGF β1作用下 ,瘢痕成纤维细胞FN及其受体α5β1整合素表达的变化。结果 TGF β1浓度在 10~ 5 0μg/L之间时 ,可明显刺激瘢痕成纤维细胞FN及其受体α5β1整合素的表达 ,与对照组相比较 ,两者差异有显著性意义 (P <0 .0 5 ) ,其作用呈一定的剂量效应关系 ,随着TGF β1浓度增加 ,促合成作用也随之增加 ,浓度在 10 0 μg/L时 ,作用达到饱和。 结论 TGF β1作用下FN及其受体α5β1整合素在成纤维细胞中的过度表达可能与增生性瘢痕形成有关。 Objective To investigate the effects of transforming growth factor-β1 (TGF-β 1) on the synthesis of FN and α 5β 1 integrin b y cultured human fibroblast isolated from hypertrophic scars. Methods The influence of TGF-β 1 at concentration of 0, 5, 10, 25, 50 and 100 μg/L on the synthesis of FN in dosage was observed using ELISA, an d influence of TGF-β 1 on the synthesis of α 5β 1 integrin was assessed b y immunohistocytochemistry and image analysis. Results TG F-β 1 could stimulate the human fibroblasts isolated from hypertrophic scars to synthesize FN andα 5β 1 integrin with dose-dependent increase at concent ration of 10~50 μg/L and the effects TGF-β 1 on the synthesis was the stron g est at concentration of 100 μg/L. Conclusions TGF-β 1 may exert its function on the formation of hypertrophic scars by stimulating th e synthesis of FN and α 5β 1 integrin.
