不同培养条件对‘丰香’草莓离体叶片再生的影响 被引量:28

摘要 以草莓品种‘丰香’离体叶片为外植体 ,探讨了基本培养基、不同细胞分裂素、暗培养、硝酸银浓度以及不同植物生长调节剂组合对不定芽再生的影响。结果表明 ,基本培养基中以MS最为适合 ,WPM、QL、AS培养基均不利于不定芽的再生 ,而TDZ的诱导效果好于BA。以MS基本培养基附加TDZ2 0mg·L-1 和IBA 0 8mg·L-1 可以使‘丰香’叶片不定芽的再生率高达 72 33%,平均每叶再生芽 5 5 9个。暗培养 1 4d可以将‘丰香’叶片的不定芽再生率提高到 90 0 9%。硝酸银对于提高‘丰香’叶片的不定芽再生没有明显效果 ,但在一定程度上改变了细胞分化的方向。 The ideal regeneration system of strawberry(Fragaria×ananassa Duch.)‘Toyonoka’ by leaf explants was established. In this study, the effects of plant growth regulators, dark periods and AgNO_3 concentrations on shoot regeneration from leaves of strawberry were investigated. The results showed that the highest regeneration rate of 72.33%, with 5.59 shoots per leaf disc, was obtained on the medium MS supplemented with 2.0 mg·L -1 TDZ and 0.8 mg·L -1 IBA. TDZ was more effective than BA in inducing shoot regeneration from leaves of strawberry‘Toyonoka’. Two weeks of dark treatment could increase the shoot regeneration rate to 90.09%. Addition of AgNO_3 to the medium could not stimulate shoot regeneration, but it changed the cell differentiational direction in somewhat from the formation of adventitious shoots to that of somatic embryos.
